The hypothesis that YB-1 activated the HGF/c-Met signaling pathway by up-regulating MACC1 also is supported by the observations that inhibition of YB-1 suppressed the expression of c-Met protein by Western blot assay (Figure (Figure4G).4G). Thus, these data provided evidence that YB-1 promoted MACC1 transcription by binding to the Y-box of the MACC1 promoter and thereby activating the MACC1/c-Met pathway.
