The pattern of glycosyl linkages and sequence of residues were determined by HMBC and ROESY (Fig. 4A). The 3-linkage was confirmed by the correlations from H-1 to C-3 in HMBC and the correlations from H-1 to H-3 in ROESY. Taking fraction II as an example, in its HMBC spectrum, a correlation from H-1 of residue B to C-3 of residue A’, H-1 of residue B to C-3 of residue A, a correlation from H-1 of residue C to C-3 of residue B, and a correlation from H-1 of residue D to C-3 of residue C were observed. In ROESY spectrum (Fig. 4A), a correlation from H-1 of residue B to H-3 of residue A0, H-1 of residue B to H-3 of residue A, a correlation from H-1 of residue C to H-3 of residue B, and a correlation from H-1 of residue D to H-3 of residue C were observed. Combining these results, the sequence of residues in fraction II is D1 ? 3C1 ? 3B1 ? 3A/A0. Similar procedures were applied to elucidate the structure of fractions I, III and IV. The sequences of residues in these oligosaccharides are C1→3B1→3A/A’, G1→3F1→3E1→3D1→3C1→3B1→3A/A’ and H1→3G1→3F1→3E1→3D1→3C1→3B1→3A/A’.
