[Abstract] Objective: to use psbA-trnH and the internal transcribed spacer (internal 2 transcribed 2 spacer, ITS2) sequence for molecular identification of fern flora. Methods: polymerase chain reaction to this kind of plant leaves and herbs (Polymerase Chain, Reaction, PCR) psbA-trnH, ITS2 amplification and sequencing, sequence and download such plant samples from the GenBank. All sequences were analyzed by DNAMAN 8 software, and the data were calculated by MEGA 6, and the neighbor-joining (NJ) was established. Results: from the phylogenetic tree analysis shows that, for all samples poly into two major branches, clubmosses Selaginella genus species were clustered into one branch, fern poly is a big branch, of which the adderstongues subclass of plant B.strictum monomer as a, with water keel plant subclass distinct, the bootstrap support to rate ideal. Conclusion: trnH intergenic region of ferns identification efficiency was 87.03%, ITS2 sequences did not get effective sequence; trnH intergenic region and the ITS2 sequence of lycopods Selaginella genus identification efficiency was 100%; therefore it can trnH intergenic region of Pteridophyte Flora in plant identification is proved to be applicable.[keyword] pteridoflora; psbA-trnH; ITS2; cluster analysis
